Dermatomyositis (DM) is a chronic, systemic autoimmune disease affecting the skin, muscle, and lungs. The activation of CB2R has been shown to reduce several, key proinflammatory cytokines implicated in DM. Our group previously that expression was significantly increased DM skin compared blood, possibly due production local inflammatory cytokines. stimulation alleviates response by down-regulating TNF-α, IL31, IFN-γ, type I interferon early stage inflammation, reducing infiltrated cells. Having more on cells will lead enhanced effects agonist. We hypothesized fibroblasts (FBs) may play an important role upregulating within skin. Therefore, we sought examine FBs activation. PBMCs, which typically have minimal expression, were cultured with human FBs. then utilized multiplexed flow cytometry further analyze 12 cell lineages. When comparing PBMCs without FBs, had significant increase CD4+ T (p<0.0001), monocyte-derived dendritic (moDCs) classical Monocytes (CM) (p<0.001), M1 Macrophages (p <0.0001). There also such as IFNβ, IFNγ, ΝFα, IL31 IL4 cells, moDCs, CD11c+ myeloid CM CD68+ macrophages.
